چکیده

Insulin release from the beta cells is influenced by nutrients (both carbohydrates and noncarbohydrates), hormonal factors including gut hormones and neural factors. Of all these insulin secretagogues, glucose availability is the major physiological determinant of insulin secretion. The insulin secretory response is greater after oral administration of glucose than after intravenous glucose administration (Fig – 1), an indication that absorption of glucose by way of gastro-intestinal tract stimulates the release of hormones and other mechanism that ultimately enhance the sensitivity of the beta cell to glucose. This phenomenon is known as ‘Incretin effect’ and is facilitated by gut hormones.

Optimization of the native glucagon sequence for medicinal purposes.

BACKGROUND Glucagon is a life-saving medication used in the treatment of hypoglycemia. It possesses poor solubility in aqueous buffers at or near physiological pH values. At low and high pH, at which the peptide can be formulated to concentrations of a milligram or more per milliliter, the chemical integrity of the hormone is limited, as evidenced by the formation of multiple degradation-relate...

The nature, significance, and glucagon-like peptide-1 analog treatment of brain insulin resistance in Alzheimer's disease.

Alzheimer's disease (AD) is an age-related neurodegenerative disease leading over the course of decades to the most common form of dementia. Many of its pathologic features and cognitive deficits may be due in part to brain insulin resistance recently demonstrated in the insulin receptor→insulin receptor substrate-1 (IRS-1) signaling pathway. The proximal cause of such resistance in AD dementia...

Clinical Application of Glucagon-Like Peptide 1 Receptor Agonists for the Treatment of Type 2 Diabetes Mellitus

Glucagon-like peptide 1 (GLP-1) is secreted from enteroendocrine L-cells in response to oral nutrient intake and elicits glucose-stimulated insulin secretion while suppressing glucagon secretion. It also slows gastric emptying, which contributes to decreased postprandial glycemic excursions. In the 1990s, chronic subcutaneous infusion of GLP-1 was found to lower blood glucose levels in patients...

Structure-activity studies of glucagon-like peptide-1.

A series of analogs of glucagon-like peptide-1 (GLP-1) was made replacing each amino acid with L-alanine to identify side-chain functional groups required for interaction with the GLP-1 receptor. In the case of L-alanine being the parent amino acid, substitution was made with the amino acid found in the corresponding position in glucagon. Binding assays were performed using the cloned rat GLP-1...

A Review on the Association between Glucagon-Like Peptide-1 Receptor Agonists and Thyroid Cancer

There is a concern on the risk of thyroid cancer associated with glucagon-like peptide-1 (GLP-1) analogs including liraglutide and exenatide. In this article, we review related experimental studies, clinical trials and observational human studies currently available. In rodents, liraglutide activated the GLP-1 receptors on C-cells, causing an increased incidence of C-cell neoplasia. Animal expe...

Glucagon-like peptide-1 analogs against antipsychotic-induced weight gain: potential physiological benefits

BACKGROUND Antipsychotic-induced weight gain constitutes a major unresolved clinical problem which may ultimately be associated with reducing life expectancy by 25 years. Overweight is associated with brain deterioration, cognitive decline and poor quality of life, factors which are already compromised in normal weight patients with schizophrenia. Here we outline the current strategies against a...
